Samples lysed at pH 8.0 in XL buffer, kept native (N) or denatured (D) with 7M urea and sonication followed by dilution to 3M Urea. Labelled with branched PEG. Quenched, native buffer normalised to urea, and reduction alkylation with 20mM IAA and varying TCEP: 10 mM (L), 20 mM (M), 40 mM (H). Cleanup via TCA acetone. Run is 0.5 ug from 3 replicates of each condition. |
